Enzyme Engineering for More Efficient Cofactor Recycling

  • This presentation we will compare the use of glucose vs. iPrOH for the regeneration of NAD(P)H cofactor on biocatalysed redox reactions.
  • Moving from glucose to the more atom efficient iPrOH requires ADHs that are very active, stable and amenable to both NADH and NADPH. We will demonstrate how we have achieved this through enzyme engineering, following rational approaches to library design.
  • For those occasions when glucose is preferable, we have designed GDH variants that can accept both cofactors while remaining active and stable.
